Utilising AI to accelerate scientific productivity will support the ability of OECD countries to grow, innovate and meet global challenges, from climate change to new contagions. This publication is aimed at a broad readership, including policy makers, the public, and stakeholders in all areas of science. Based on apical-basal and planar cell polarities’ ubiquity, I suggest a 3D mathematical model: Point particles represent cells having zero, one, or two unit-arrows representing polarities. I test the model abilities on preimplantation development, sea urchin gastrulation, mammalian neurulation, organoid folding, and tubulogenesis. I find that a minimal, versatile toolbox, including cellular polarities, captures the emergence of diverse and robust animal morphologies. Part 2: How are deep convective events spatially organized in the tropical atmosphere? Here, I test the importance of atmospheric cold pools for organizing convection. I suggest a 2D mathematical model: Points expand into circles representing cold pools. When circles meet, a convective event occurs, and a new circle forms. I find this model captures convective scale increase and initial stages of convective self-aggregation. The latter is crucial due to its link to tropical cyclogenesis.

McCabe, 2022-06-09 This open access volume compiles student reports from the 2021 Simula Summer School in Computational Physiology. Interested readers will find herein a number of modern approaches to modeling excitable tissue. This should provide a framework for tools available to model subcellular and tissue-level physiology across scales and scientific questions. In June through August of 2021, Simula held the seventh annual Summer School in Computational Physiology in collaboration with the University of Oslo (UiO) and the University of California, San Diego (UCSD). The course focuses on modeling excitable tissues, with a special interest in cardiac physiology and neuroscience. The majority of the school consists of group research projects conducted by Masters and PhD students from around the world, and advised by scientists at Simula, UiO and UCSD.   npj systems biology and applications impact factor: Pharmacogenetic biomarkers for chemotherapy-induced adverse drug reactions Niclas Björn, 2019-11-20 Cancer is a serious disease expected to be the world-leading cause of death in the 21st century. The use of harsh chemotherapies is motivated and accepted but, unfortunately, is often accompanied by severe toxicity and adverse drug reactions (ADRs). These occur because the classical chemotherapies’ common modes of action effectively kill and/or reduce the growth rate not only of tumour cells, but also of many other rapidly dividing healthy cells in the body. There are also considerable interindividual differences in ADRs, even between patients with similar cancers and disease stage treated with equal doses; some experience severe to life-threatening ADRs after one dose, leading to treatment delays, adjustments, or even discontinuation resulting in suboptimal treatment, while others remain unaffected through all treatment cycles. Being able to predict which patients are at high or low risk of ADRs, and to adjust doses accordingly before treatment, would probably decrease toxicity and patient suffering while also increasing treatment tolerability and effects. In this thesis, we have used next-generation sequencing (NGS) and bioinformatics for the prediction of myelosuppressive ADRs in lung and ovarian cancer patients treated with gemcitabine/carboplatin and paclitaxel/carboplatin. Paper I shows that ABCB1 and CYP2C8 genotypes have small effects inadequate for stratification of paclitaxel/carboplatin toxicity. This supports the transition to whole-exome sequencing (WES) and whole-genome sequencing (WGS). Papers II and IV, respectively, use WES and WGS, and demonstrate that genetic variation in or around genes involved in blood cell regulation and proliferation, or genes differentially expressed at chemotherapy exposure, can be used in polygenic prediction models for stratification of gemcitabine/carboplatininduced myelosuppression. Paper III reassuringly shows that WES and WGS are concordant and mostly yield comparable genotypes across the exome. Paper V proves that single-cell RNA sequencing of hematopoietic stem cells is a feasible method for elucidating differential transcriptional effects induced as a response to in vitro chemotherapy treatment. In conclusion, our results supports the transition to genome-wide approaches using WES, WGS, and RNA sequencing to establish polygenic models that combine effects of multiple pharmacogenetic biomarkers for predicting chemotherapy-induced ADRs. This approach could be applied to improve risk stratification and our understanding of toxicity and ADRs related to other drugs and diseases. We hope that our myelosuppression prediction models can be refined and validated to facilitate personalized treatments, leading to increased patient wellbeing and quality of life.

Essential oils from medicinal and aromatic plants (MAPs) have been exploited for product formulations of pharmaceuticals, cosmetics, food and beverage, colorants, biopesticides, and several other utility chemicals of industrial importance. There is scientific evidence of many medicinal plant extracts possessing immunomodulatory, immunostimulatory, antidiabetic, anticarcinogenic, antimicrobial, and antioxidant properties, thus demonstrating their traditional use in popular medicine. With the advent of modern technology, the exploitation of natural resources has exponentially increased in order to fulfill the demand of an increased human population with improved quality of life. The traditional agriculture and production-based supply of commodities is inadequate to meet the current demand. Biotechnological approaches are gaining importance to bridge the gaps in demand and supply. In the proposed book, medicinal and aromatic plant-based secondary metabolites have been discussed in terms of their therapeutic potential and industrial relevance. To discuss the qualitative and quantitative analysis of a range of medicinal and aromatic plants-based secondary metabolites (SMs), bioprocess development for their extraction and bioseparation, a brief overview of their industrial relevance, various tissue culturing strategies, biotechnological approaches to enhance production, scale-up strategies, management of residual biomass post extraction of target SMs is central to the idea of the proposed book. A section will explore the verticals mentioned above. In the next section, the book addresses the approaches for conserving and improving medicinal and aromatic plant genetic resources. In the third section, approaches to managing the post-harvest crop residue and secondary metabolites extracted plant biomass will be thoroughly discussed. The recent integration of artificial intelligence to improve medicinal and aromatic plant research at several levels, including the development and employment of computational approaches to enhance secondary metabolite production, tissue culture, drug design and discovery, and disease treatment, will be included in the fourth section. The book summarizes current research status, gaps in knowledge, agro-industrial potential, waste or residual plant biomass management, conservation strategies, and computational approaches in the area of medicinal and aromatic plants with an aim to translate biotechnological interventions into reality.
