  ado roi: Long-Term Consequences of Adolescent Drug Use: Evidence from Pre-Clinical and Clinical Models Mary M. Torregrossa, Jacqueline M. Barker, Shannon L. Gourley, 2018-09-05 The purpose of this collection is to provide a forum to integrate pre-clinical and clinical investigations regarding the long-term consequences of adolescent exposure to drugs of abuse. Adolescence is characterized by numerous behavioral and biological changes, including substantial neurodevelopment. Behaviorally, adolescents are more likely to engage in risky activities and make impulsive decisions. As such, the majority of substance use begins in adolescence, and an earlier age of onset of use (<15 yr) is strongly associated with the risk for developing a substance use disorder later in life. Furthermore, adolescent drug use may negatively impact ongoing neurological development, which could lead to long-term cognitive and emotional deficits. A large number of clinical studies have investigated both the acute and long-term effects of adolescent drug use on functional outcomes. However, the clinical literature contains many conflicting findings, and is often hampered by the inability to know if functional differences existed prior to drug use. Moreover, in human populations it is often very difficult to control for the numerous types of drugs, doses, and combinations used, not to mention the many other environmental factors that may influence adult behavior. Therefore, an increase in the number of carefully controlled studies using relevant animal models has the potential to clarify which adolescent experiences, particularly what drugs used when, have long-term negative consequences. Despite the advantages of animal model systems in clarifying these issues, the majority of pre-clinical addiction research over the past 50+ years has been conducted in adult animals. Moreover, few addiction-related studies have investigated the long-term neurocognitive consequences of drug exposure at any age. In the past 10 years of so, however, the field of adolescent drug abuse research has burgeoned. To date, the majority of this research has focused on adolescent alcohol exposure using a variety of animal models. The results have given the field important insight into why adolescents are more likely to drink alcohol to excess relative to adults, and the danger of adolescent alcohol use (e.g., in leading to a persistence of excessive drinking in adulthood). More recently, research regarding the effects of adolescent exposure to other drugs of abuse, including nicotine, cocaine, and cannabinoids has expanded. Therefore, we are at unique point in time, when emerging results from carefully controlled pre-clinical studies can inform the sometimes confusing clinical literature. In addition, we expect an influx of prospective clinical studies in response to a cross-institute initiative at NIH, known as the ABCD grant. Several institutes are enrolling children prior to adolescence (and the initiation of drug use), in order to control for pre-existing neurobiological and neurobehavioral differences and to monitor the age of initiation and amount of drug used more carefully than is possible using retrospective designs.

Some of the 24 submissions were merged and resulted in the 21 papers that are presented in this book. The papers explore methods for predicting fluid intelligence from T1-weighed MRI of 8669 children (age 9-10 years) recruited by the Adolescent Brain Cognitive Development Study (ABCD) study; the largest long-term study of brain development and child health in the United States to date.

Please see the first volume here. Binge drinking (BD), also known as heavy episodic drinking or college drinking, is a highly prevalent pattern in most Western countries characterized by the intake of large amounts of alcohol in a short time followed by periods of abstinence. This excessive pattern of alcohol use is a regular practice in around a third of European and American youth. The high prevalence of BD at this age is of particular concern since adolescents are in a period of special vulnerability to neurotoxic effects of alcohol, mainly due to the structural and functional changes going on in the brain throughout this key developmental stage. As showed in the first volume of the Research Topic, evidence gathered during the last decade from animal and human studies seems to point out multiple brain anomalies associated with BD at different levels: biochemical, structural, functional, but also cognitive and affective. In this Second Volume, our aim is to bring together the most recent studies –namely original research and systematic or critical reviews- which provide a comprehensive approach regarding the effects of BD on the adolescent and young brain. Thus, we aim to include manuscripts from different domains (preclinical and clinical research), perspectives (genetic, behavioral, neuropsychological, clinical psychology, psychophysiology, neuroimaging, etc.), and methods (behavioural, biochemical, EEG, MEG, MRI, etc.). Likewise, we welcome submissions aimed at examining the effects of BD beyond the brain (personality dimensions, polygenic risk, peripheral inflammation, cortisol response, gut microbiome, etc.) and approaches focused on reducing BD, such as cognitive training and/or neuromodulation (by tDCS, TMS, etc.).

The high prevalence of BD at this age is of particular concern since adolescence and youth are in a period of special vulnerability to neurotoxic effects of alcohol, mainly due to the structural and functional changes going on in the brain throughout this key developmental stage. Evidence gathered during the last decade from animal and human studies seems to point to multiple brain anomalies associated with BD. In this Research Topic, we have collated a compendium of articles that address multiple aspects of BD during adolescence and young adulthood such as identification, prevalence, gender differences and neurocognitive anomalies associated with this excessive alcohol consumption pattern. These articles collectively highlight the breadth of current research conducted in this field but also the need to join efforts to improve the screening of the BD pattern, the characterization of its consequences as well as the translation of knowledge acquired in the laboratory into clinical practice. We remain confident that this Research Topic will contribute significantly to the understanding of BD and its consequences and will further stimulate high-quality investigation in this relatively new research field.

  ado roi: High-Level Models of Unconventional Computations Andrew Schumann, Krzysztof Pancerz, 2018-05-17 This book shows that the plasmodium of Physarum polycephalum can be considered a natural labelled transition system, and based on this, it proposes high-level programming models for controlling the plasmodium behaviour. The presented programming is a form of pure behaviourism: the authors consider the possibility of simulating all basic stimulus–reaction relations. As plasmodium is a good experimental medium for behaviouristic models, the book applies the programming tools for modelling plasmodia as unconventional computers in different behavioural sciences based on studying the stimulus–reaction relations. The authors examine these relations within the framework of a bio-inspired game theory on plasmodia they have developed i.e. within an experimental game theory, where, on the one hand, all basic definitions are verified in experiments with Physarum polycephalum and Badhamia utricularis and, on the other hand, all basic algorithms are implemented in the object-oriented language for simulations of plasmodia. The results allow the authors to propose that the plasmodium can be a model for concurrent games and context-based games.
